CHARM Therapeutics Doses First AML Patient in CHM-029 Trial
CHARM Therapeutics has dosed its first patient and fully enrolled the opening cohort in a Phase I/II trial of CHM-029 for acute myeloid leukemia.
CHARM Therapeutics announced Wednesday that it has dosed the first patient and completed enrollment of the initial cohort in a Phase I/II clinical trial of CHM-029, its lead drug candidate targeting acute myeloid leukemia (AML). The London-based clinical-stage biotechnology company described the milestone as a significant step forward in its effort to address drug resistance in one of the most aggressive blood cancers.
CHM-029 is a menin inhibitor that CHARM characterizes as best-in-class. Menin inhibition has emerged as a clinically validated approach in AML, a disease where resistance to existing therapies remains a central challenge and a cause of poor long-term outcomes for many patients.

The compound carries an additional distinction that sets it apart in the broader drug-development landscape: CHM-029 is the first molecule designed using a protein-ligand co-folding computational model to advance into human clinical studies. The approach applies AI-driven structural prediction to the drug design process, reflecting growing industry reliance on machine learning tools to accelerate early-stage discovery.
The Phase I/II trial structure allows investigators to assess safety and tolerability in early cohorts before expanding to evaluate efficacy signals at higher dose levels. Full enrollment of the first cohort marks the completion of an initial gating requirement before the study can progress to subsequent stages.
